**CodaVax-DENV** is a next-generation, tetravalent, live-attenuated dengue vaccine candidate developed by Codagenix Inc. using a proprietary synthetic attenuated virus engineering (SAVE) platform that employs codon deoptimization to rationally attenuate contemporary strains of all four dengue virus (DENV) serotypes. This approach introduces hundreds of strategic genomic modifications to slow viral replication, creating a genetically stable virus incapable of reverting to wild-type while preserving all viral proteins as antigens to elicit robust systemic and mucosal immunity, including neutralizing antibodies and cellular T-cell responses that mimic natural infection. Designed for balanced immunogenicity across serotypes without the antibody-dependent enhancement (ADE) risks seen in some prior dengue vaccines, it is supported by U.S. Department of Defense funding and aims to address dengue prevention in endemic regions.[3][7][8][9]
