Overview

**Codrug 1** is an experimental, lipophilic small-molecule mutual prodrug created at the University \"G. d'Annunzio\" of Chieti-Pescara by covalently linking **(R)-alpha-lipoic acid** and **ibuprofen** through a diamide linkage. It was designed to improve brain delivery of the anti-inflammatory and antioxidant parent drugs in neurodegenerative disease. In rat beta-amyloid 1-40 infusion models of Alzheimer's disease, codrug 1 showed slow conversion to ibuprofen and (R)-alpha-lipoic acid, antioxidant activity, and greater protection than ibuprofen against amyloid-associated cognitive, behavioral, biochemical, and morphological abnormalities. It is a preclinical research compound, has no assigned INN or marketed trade name, and is not an approved medicine.
