COF-01 refers to a novel therapeutic target and the associated Raptamer-drug conjugate (Rap-DC) program developed by Fannin Innovation Studio in collaboration with Champions Oncology. The therapeutic candidate utilizes a next-generation base-modified single-stranded DNA aptamer (Raptamer) to target the COF-01 protein, which was identified through proteomics and immunohistochemistry as being overexpressed in several solid tumors, including non-small cell lung cancer (NSCLC), head and neck cancer, and pancreatic cancer. The Raptamer is conjugated to the cytotoxic payload monomethyl auristatin F (MMAF) via a cleavable linker. This modality is designed to offer superior tumor penetration compared to traditional antibody-drug conjugates (ADCs) due to its smaller molecular size, while maintaining high specificity and a favorable safety profile. Preclinical studies have demonstrated that COF-01 Rap-DCs selectively induce cytotoxicity in COF-01-positive human cancer cells, correlating with poor overall survival in patients with high COF-01 expression.
