Drug intelligence / Profile preview
COF-01-016 is a preclinical-stage Raptamer-drug conjugate (Rap-DC) being developed by Raptamer Discovery Group (a Fannin portfolio company) in collaboration with Champions Oncology for the treatment of solid tumors, including non-small cell lung cancer (NSCLC). The therapeutic candidate consists of a proprietary, base-modified single-stranded DNA (ssDNA) aptamer (Raptamer) that targets COF-01, a novel tumor-associated antigen identified through proteomic analysis of patient-derived tumor samples. The Raptamer is conjugated via a cleavable linker to a potent cytotoxic payload, such as monomethyl auristatin E (MMAE) or monomethyl auristatin F (MMAF). Upon binding to COF-01 on the surface of cancer cells, the conjugate is internalized, releasing the payload to inhibit tubulin polymerization and selectively induce cell death in COF-01-positive malignancies while sparing normal tissues.
