**COF-Rapa** is an experimental, sustained-release intravitreal nanomedicine consisting of rapamycin loaded into a water-dispersible covalent organic framework nanoparticle carrier. In a mouse retinal ischemia-reperfusion model intended to mimic acute glaucoma-associated retinal injury, a single intravitreal injection produced prolonged retinal inhibition of mechanistic target of rapamycin complex 1 for up to six weeks, reduced retinal inflammation, and enhanced retinal ganglion cell protection relative to the unloaded covalent organic framework carrier. It is a preclinical ophthalmic delivery formulation and has not been identified as a marketed product or clinical-stage program.
