COF008 is a humanized monoclonal antibody targeting Cluster of Differentiation 24 (CD24), a glycosylphosphatidylinositol (GPI)-anchored membrane protein that serves as a potent "don't eat me" signal in the tumor microenvironment. Developed by Lee's Pharmaceutical through its oncology-focused subsidiary, China Oncology Focus Limited (COF), COF008 is designed to disrupt the interaction between CD24 on tumor cells and Sialic acid-binding Ig-like lectin 10 (Siglec-10) on macrophages. This interaction normally inhibits phagocytosis, allowing cancer cells to evade immune surveillance. By blocking this innate immune checkpoint, COF008 promotes the phagocytic clearance of malignant cells by macrophages. It is currently being investigated in early-stage clinical trials for the treatment of advanced solid tumors.
