COF009 is a small molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1) currently in preclinical development by Lee's Pharmaceutical and its subsidiary ZKGZ Biotech (Zhaoke Guangzhou). HPK1 is a member of the MAP4K family and functions as a negative regulator of T-cell receptor (TCR) signaling. By inhibiting HPK1, COF009 is designed to augment T-cell activation and proliferation, thereby reversing immunosuppression in the tumor microenvironment and enhancing the immune system's ability to recognize and destroy tumor cells. The drug is being evaluated for the treatment of various solid tumors as a potential immuno-oncology therapy.
