COF010 is a preclinical-stage small molecule inhibitor of Ectonucleotide Pyrophosphatase/Phosphodiesterase 1 (ENPP1), being developed by Lee's Pharmaceutical through its oncology-focused subsidiary, China Oncology Focus (COF). ENPP1 is an enzyme responsible for the degradation of cyclic GMP-AMP (cGAMP), which is a critical endogenous activator of the Stimulator of Interferon Genes (STING) pathway. By inhibiting ENPP1, COF010 prevents the breakdown of cGAMP, thereby promoting the activation of the STING pathway, which triggers the production of Type I interferons and other pro-inflammatory cytokines. This mechanism is intended to enhance the body's innate immune response against tumors and improve the infiltration of T cells into the tumor microenvironment. COF010 is currently being evaluated for the treatment of various solid tumors.
