COG-301 is an investigational small interfering RNA (siRNA) therapeutic being developed by Cognigenics for the treatment of Parkinson's disease psychosis (PDP) and associated anxiety. The drug is formulated within lipid nanoparticles (LNPs) and is designed for intranasal administration, a delivery route intended to bypass the blood-brain barrier by utilizing the olfactory and trigeminal nerve pathways for direct-to-brain transport. COG-301 functions by knocking down the expression of the 5-HT2A receptor (HTR2A) at the mRNA level. By reducing the density of these G-protein coupled receptors (GPCRs) in specific brain regions, the therapy aims to mitigate psychotic symptoms and anxiety while potentially avoiding the systemic side effects and metabolic complications associated with traditional oral antipsychotics. COG-301 is currently in the preclinical stage of development.
