COG112 is an investigational apolipoprotein E (apoE)-mimetic therapeutic peptide, consisting of an apoE receptor‑binding domain fused to an antennapedia cell‑penetrating sequence, being developed by Cognosci for inflammatory and neurodegenerative indications including multiple sclerosis, traumatic and ischemic CNS injury, diabetic peripheral neuropathy and inflammatory bowel disease. Preclinical studies demonstrate that COG112 inhibits NF‑κB signaling and IκB kinase activity, reduces expression of inflammatory mediators such as iNOS, TNF‑α, IFN‑γ, IL‑17 and chemokines, and attenuates neuroinflammation and colitis in animal models, while also promoting axonal regeneration, remyelination and functional recovery in models of experimental autoimmune encephalomyelitis, sciatic nerve crush, traumatic brain injury and other neurological disorders.[1][4][10][11] In Alzheimer’s‑related models, COG112 reduces neuroinflammation, protects adult hippocampal neurogenesis and mitigates tau hyperphosphorylation and somatodendritic accumulation, supporting its potential as a disease‑modifying neurorestorative agent.[5][11]
