COG1410 is a novel, small synthetic apolipoprotein E (apoE)-mimetic peptide developed by Cognosci, Inc. for the treatment of traumatic brain injury (TBI) and Alzheimer's disease. Composed of apoE residues 138-149 with aminoisobutyric acid (Aib) substitutions at positions 140 and 145, the peptide mimics the receptor-binding region of the multifunctional apoE protein. COG1410 exhibits potent anti-inflammatory and neuroprotective activities, including the suppression of matrix metalloproteinase-9 (MMP-9) activity and the reduction of blood-brain barrier disruption. In preclinical models, it has demonstrated the ability to improve motor and cognitive functions, reduce lesion volume and vasogenic edema in TBI, and decrease amyloid-beta deposition in Alzheimer's models.
