COH06 is a genetically engineered natural killer (NK) cell therapy developed for the treatment of advanced non-small cell lung cancer (NSCLC) that is refractory to prior PD-1/PD-L1 immune checkpoint inhibitor therapy. The NK cells used in COH06 are derived from umbilical cord blood and are modified to express soluble interleukin-15 (sIL-15) and programmed death-ligand 1 (PD-L1). The expression of sIL-15 enhances NK cell survival and proliferation, while PD-L1 expression may increase the cytotoxic activity of these cells against tumor cells. This approach aims to improve anti-tumor immunity in patients whose cancers have not responded to standard immunotherapies. COH06 is being evaluated as monotherapy or in combination with atezolizumab, a monoclonal antibody targeting PD-L1, in phase 1 clinical trials[1][2][3][5].
