COH29 is a potent ribonucleotide reductase (RNR) inhibitor with anticancer activity. It works by binding to the ligand-binding pocket of the RNR M2 subunit (hRRM2) near the C-terminal tail, blocking the interaction between hRRM1 and hRRM2 subunits and interfering with the assembly of the active RNR complex. Developed at City of Hope Cancer Center, COH29 has shown efficacy against multiple cancer types in preclinical studies, particularly leukemia and ovarian cancer.
