Coixol is a natural benzoxazolinone compound primarily extracted from the plant *Coix lacryma-jobi L.* (Job's tears). It is recognized for its significant anti-inflammatory, antioxidant, and neuroprotective properties. In preclinical research, particularly in models of Parkinson's disease, coixol has demonstrated the ability to ameliorate dopaminergic neurodegeneration by inhibiting neuroinflammation and maintaining mitochondrial integrity. Its mechanism of action involves the suppression of the nuclear factor kappa B (NF-κB) and mitogen-activated protein kinase (MAPK) signaling pathways, as well as the regulation of the NLRP3/Caspase-1/IL-1β inflammasome axis. By reducing the production of reactive oxygen species (ROS), coixol prevents mitochondrial damage and subsequent inflammasome overactivation, thereby protecting neurons from inflammatory damage.
