COL-1 is a murine IgG2a monoclonal antibody with high affinity for carcinoembryonic antigen (CEA), a tumor-associated glycoprotein commonly overexpressed in various cancers, particularly colorectal cancer. This antibody does not react with CEA-related antigens such as nonspecific cross-reacting antigen (NCA) or normal fecal antigen (NFA). In preclinical studies, radiolabeled forms of COL-1 (e.g., 125I-COL-1 and 131I-COL-1) have demonstrated efficient and specific targeting of human colon carcinoma xenografts in athymic mice. Therapeutically, administration of radiolabeled COL-1 has shown reduction in tumor growth rates, especially when delivered using dose fractionation protocols to minimize toxicity and maximize efficacy. These findings support the potential use of radiolabeled COL-1 as a targeted radioimmunotherapy agent for CEA-expressing tumors[6].
