COL01 CAR-T is a second-generation chimeric antigen receptor (CAR) T-cell therapy targeting CD19, developed by the Instituto Distrital de Ciencia, Biotecnología e Innovación en Salud (IDCBIS) in Bogotá, Colombia. The construct features a CD19-specific single-chain variable fragment (scFv), a CD8 transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain, delivered via a third-generation lentiviral vector. COL01 is being investigated for the treatment of B-cell malignancies, with research focusing on optimizing manufacturing efficiency. Studies have compared a standard 10-day automated protocol using the CliniMACS Prodigy platform with a 3-day semi-automated "fast" protocol. Preclinical data in NSG-NALM6 xenograft models indicate that the fast-protocol product (COL01-CAR-TFAST) achieves higher transduction efficiency, enhanced activation phenotypes, and superior in vivo persistence and antileukemic activity compared to standard automated manufacturing methods.
