COL1A1 shRNA is a gene-silencing therapeutic candidate designed to downregulate the expression of Collagen Type I Alpha 1 (COL1A1). In glioblastoma (GBM), COL1A1 is a critical component of the extracellular matrix (ECM) that is highly expressed in mesenchymal-like glioma cells. These cells form organized structures known as oncostreams, which facilitate tumor cell migration and invasion. By utilizing short hairpin RNA (shRNA) to deplete COL1A1, this approach aims to disrupt oncostream formation, alter the recruitment and activation of reactive astrocytes and immunosuppressive macrophages, and inhibit vascular proliferation. Preclinical research at the University of Alabama at Birmingham has demonstrated that targeting the COL1A1-mediated ECM-astrocyte interaction can significantly impact the brain microenvironment and reduce tumor aggression in orthotopic mouse models.
