COL2A1 antibody is an experimental monoclonal antibody designed to target type II collagen alpha 1 (COL2A1), a protein that is highly expressed in the extracellular matrix (ECM) of high-risk Wilms tumors but absent in normal kidney tissue. Developed by researchers at Children's Hospital Los Angeles and the University of Southern California, the antibody works by blocking COL2A1-mediated signaling, which otherwise activates the AKT pathway, induces epithelial-mesenchymal transition (EMT), and promotes chemoresistance. In preclinical studies using patient-derived xenografts (PDXs), treatment with the COL2A1 antibody has been shown to reverse EMT, restore the expression of tumor suppressors, and significantly enhance the sensitivity of tumor cells to chemotherapy, offering a potential new strategy for treating pediatric renal malignancies.
