Overview

**Colchicine + tocilizumab** is a combination of two anti-inflammatory agents being investigated for the treatment of severe COVID-19 pneumonia and cytokine storm syndrome. Colchicine is an oral small molecule that acts by inhibiting the NLRP3 inflammasome, thereby suppressing the early phase of the cytokine cascade and reducing interleukin-1 (IL-1) and interleukin-6 (IL-6) production. Tocilizumab is a recombinant humanized monoclonal antibody that blocks the IL-6 receptor, inhibiting the downstream effects of IL-6-mediated pro-inflammatory signaling. The combination seeks to provide a synergistic blockade of both upstream (colchicine) and downstream (tocilizumab) inflammatory pathways in severe hyperinflammatory states such as COVID-19 pneumonia.
