Overview

This is an investigational antibiotic combination therapy consisting of colistin (colistimethate sodium) and meropenem. Colistin is a polymyxin antibiotic that disrupts the bacterial outer membrane by binding to lipopolysaccharides, while meropenem is a carbapenem that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins. The combination was evaluated by the University of Michigan in the Phase 3 OVERCOME trial for treating severe infections, including pneumonia and bloodstream infections, caused by extensively drug-resistant (XDR) and carbapenem-resistant Gram-negative bacilli such as Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacterales. Clinical trial results indicated that the addition of meropenem to colistin did not significantly improve 30-day mortality or clinical success rates compared to colistin monotherapy.
