colivelin

Colivelin is a synthetic hybrid peptide designed to enhance neuroprotection, particularly in the context of neurodegenerative diseases. It is composed of the Activity-Dependent Neurotrophic Factor (ADNF) C-terminally fused to AGA-(C8R) HNG17, a potent derivative of humanin. Colivelin acts as a brain-penetrant neuroprotective agent and has demonstrated potent activity in suppressing neuronal death induced by familial Alzheimer's disease-causing genes and beta-amyloid toxicity. Its mechanism involves activation of the JAK/STAT3 signaling pathway, specifically acting as an activator of STAT3, which contributes to its anti-apoptotic and cytoprotective effects. Preclinical studies have shown that intraperitoneal administration can suppress memory impairment in animal models and ameliorate endothelial dysfunction in sepsis by reducing inflammation and preserving vascular integrity[1][2][4][5][7][10]. Colivelin is under investigation primarily for Alzheimer's disease but has also shown potential for treating ischemic brain injury and sepsis-associated endothelial dysfunction.
