Overview

Collagenase (marketed as Santyl) is a topical enzymatic debriding agent used to remove necrotic (dead) tissue from chronic skin ulcers and severely burned areas. The active ingredient, collagenase, is an enzyme derived from the fermentation of *Clostridium histolyticum*. Its mechanism of action involves selectively breaking down and digesting denatured or damaged collagen in necrotic tissue while sparing healthy granulation tissue. This selective enzymatic debridement promotes wound healing by clearing barriers to new tissue growth and facilitating the formation of granulation tissue and subsequent re-epithelialization. Collagen accounts for about 75% of the dry weight of skin; thus, its targeted removal in nonviable tissues accelerates wound bed preparation for healing[1][2][3][6]. Collagenase Santyl Ointment is FDA-approved for use in adults with chronic dermal ulcers and severe burns[6].
