Overview
Colon GVAX is an investigational allogeneic, whole-cell cancer vaccine designed for the treatment of colorectal cancer. It consists of two colorectal cancer cell lines (SW837 and SW620) and a bystander cell line engineered to secrete granulocyte-macrophage colony-stimulating factor (GM-CSF), which acts as an adjuvant to stimulate antigen-presenting cells and induce a systemic T-cell immune response against tumor-associated antigens. The vaccine aims to enhance anti-tumor immunity in patients with mismatch repair proficient (MMRp) colorectal cancer, a population typically resistant to single-agent PD-1 inhibitor therapy. Colon GVAX has been studied in combination with low-dose cyclophosphamide (to inhibit regulatory T cells) and pembrolizumab in phase 2 clinical trials for advanced or metastatic colorectal cancer[2][3][4][5][6].
