Overview

Coltuximab ravtansine is an investigational antibody-drug conjugate composed of a chimeric monoclonal antibody targeting B-lymphocyte antigen CD19, a B-cell surface antigen, conjugated to the cytotoxic maytansinoid DM4 via a disulfide linker. The drug selectively binds to CD19 on B cells, leading to internalization and intracellular release of DM4, which inhibits tubulin polymerization and microtubule assembly. This mechanism disrupts cell division and induces apoptosis in malignant B cells. Coltuximab ravtansine was developed primarily for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and has also been investigated in precursor cell lymphoblastic leukemia/lymphoma. The drug was originally developed by ImmunoGen (now part of AbbVie), with clinical trials reaching phase II for DLBCL[1][2][3][4][5].
