Overview

COM1010 (formerly known by the discovery code CGEN-438) is a high-affinity, potential first-in-class monoclonal antibody developed by Compugen that targets the immune checkpoint protein **B7-H4** (also known as VTCN1). B7-H4 is a member of the B7 family of costimulatory molecules and is frequently overexpressed in various solid tumors, including ovarian, breast, and endometrial cancers, where it acts as a negative regulator of T-cell mediated anti-tumor immunity. By binding to B7-H4, COM1010 blocks its inhibitory signaling, thereby restoring T-cell activation, proliferation, and cytokine production within the tumor microenvironment. The drug is currently being evaluated in Phase 1 clinical trials as a monotherapy and in combination with other immunotherapies to address unmet needs in patients with advanced solid tumors that exhibit high B7-H4 expression.
