Overview

COM701 is a first-in-class, humanized IgG4 monoclonal antibody developed by Compugen. It targets the poliovirus receptor-related immunoglobulin domain containing protein (PVRIG, also known as CD112R), an immune checkpoint molecule. By binding to PVRIG and blocking its interaction with its ligand PVRL2, COM701 inhibits this immune checkpoint pathway, leading to enhanced activation of T cells and natural killer (NK) cells in the tumor microenvironment. This mechanism aims to generate antitumor immune responses and overcome resistance in tumors that are typically unresponsive to other immunotherapies. Preclinical studies have shown that blockade of PVRIG can inhibit tumor growth, and clinical trials have demonstrated preliminary antitumor activity—especially when combined with other checkpoint inhibitors such as nivolumab (anti-PD1) or BMS-986207 (anti-TIGIT). The drug is being investigated for use in various solid tumors including non-small cell lung cancer, ovarian cancer, breast cancer, endometrial cancer, and colorectal cancer[1][2][3][5][6].
