Comotaxel is a novel semi-synthetic taxane derivative developed by the Institute of Materia Medica, Chinese Academy of Medical Sciences (IMM, CAMS). Functioning as a microtubule stabilizer, comotaxel binds to the beta-subunit of tubulin, promoting microtubule assembly and inhibiting disassembly. This mechanism leads to the stabilization of the mitotic spindle, causing cell cycle arrest at the G2/M phase and ultimately inducing apoptosis in cancer cells. Comotaxel was designed to improve upon the efficacy and safety profile of first-generation taxanes like paclitaxel and docetaxel, particularly in overcoming certain resistance mechanisms. It has been investigated in clinical trials for the treatment of various advanced solid tumors, including breast cancer, non-small cell lung cancer (NSCLC), and gastric cancer, and is typically administered via intravenous infusion.
