Drug intelligence / Profile preview
Compartment-locked IL-12 (also known as InC01 or InC-01) is a preclinical-stage, compartment-locked recombinant fusion protein consisting of interleukin-12 (IL-12) cytokine fused to an IgG4 Fc domain. Developed by InCephalo Therapeutics (a spin-off from the University of Zurich), it is engineered using the proprietary C-Lock platform to abrogate binding to the neonatal Fc receptor (FcRn) through specific amino-acid substitutions at the Fc:FcRn interface. In glioblastoma, FcRn is upregulated and typically exports Fc-fused biologics from the brain into the systemic circulation, leading to peripheral toxicity. By silencing FcRn binding, compartment-locked IL-12 remains localized within the central nervous system (CNS) compartment after local administration (such as convection-enhanced delivery), enhancing local anti-tumor efficacy while minimizing systemic exposure and associated cytokine-related toxicities.
