Compound 073 is a synthetic chalcone derivative identified as a potential antiproliferative agent for the treatment of ovarian cancer. Developed through 3D-QSAR (Quantitative Structure-Activity Relationship) modeling using Comparative Molecular Field Analysis (CoMFA) and Comparative Molecular Similarity Index Analysis (CoMSIA), it was synthesized and experimentally validated against the A2780 ovarian cancer cell line. The compound exerts its cytotoxic effects via a reactive oxygen species (ROS)-dependent mechanism, which leads to the upregulation of heme oxygenase-1 (HO-1) expression. Notably, this induction of HO-1 occurs independently of the canonical Nrf2 signaling pathway. Compound 073 is part of a series of chalcone-based inhibitors designed to overcome chemoresistance, demonstrating activity in both standard and cisplatin-resistant ovarian cancer models. Its cytotoxicity can be abrogated by the antioxidant N-acetylcysteine (NAC), confirming the central role of ROS in its mode of action.
