Overview

Compound 11a is a novel, water-soluble semicarbazide-containing sulfonamide derivative developed by researchers at Zhengzhou University for the topical treatment of glaucoma. It functions as a carbonic anhydrase (CA) inhibitor, specifically targeting human carbonic anhydrase II (hCA II). The compound was designed using the "sugar tail method," incorporating a glucose moiety to enhance water solubility and achieve a neutral pH in aqueous solution, which is critical for non-irritating topical ocular administration. In preclinical studies using glaucomatous rabbit models, compound 11a demonstrated significant intraocular pressure (IOP) lowering effects when applied as a 1% or 2% solution. Furthermore, it exhibits favorable drug-like properties, including low cytotoxicity toward human corneal epithelial cells and no significant inhibition of hERG channels, suggesting a low risk of cardiotoxicity.
