Compound 15 is a small molecule, G protein-coupled receptor kinase (GRK)-biased agonist of the beta-2 adrenergic receptor (beta-2AR). Unlike traditional beta-2AR agonists that primarily activate the Gs-cAMP pathway, compound 15 is designed to selectively trigger GRK2-mediated signaling. This biased signaling mechanism promotes glucose uptake via an mTORC2-dependent pathway while minimizing cAMP-related side effects such as tachycardia and tremors. It is being investigated for the treatment of type 2 diabetes and obesity, and has shown potential in reversing muscle atrophy associated with incretin-based therapies. Clinical data indicates that the compound is bioavailable and well-tolerated in both healthy volunteers and patients with type 2 diabetes.
