Overview

Compound 15 is a potent and selective small molecule inhibitor of cyclin-dependent kinase 2 (CDK2), discovered by researchers at the University of South Australia. It belongs to the N,4-di(1H-pyrazol-4-yl)pyrimidin-2-amine chemical class, developed through bioisosteric replacement of phenylsulfonamide moieties in earlier lead compounds. In preclinical studies, Compound 15 demonstrated sub-micromolar antiproliferative activity against a panel of 13 cancer cell lines, including ovarian cancer. Its mechanism of action involves the inhibition of CDK2, which leads to reduced phosphorylation of the retinoblastoma (Rb) protein at Thr821. This inhibition results in cell cycle arrest at the S and G2/M phases and the induction of apoptosis in cancer cells. It is currently a research-stage compound intended for the treatment of various cancers.
