Compound 22a is a non-specific designation used in medicinal chemistry literature to refer to distinct experimental small molecules. One version is a sulfonyl-hydrazone derivative of dantrolene developed as a dual inhibitor of ryanodine receptors (RyR) and acetylcholinesterase (AChE). This compound is intended for the treatment of Alzheimer's disease, where it aims to restore calcium homeostasis and increase acetylcholine levels; it has demonstrated the ability to improve memory, cognitive behavior, and synaptic strength in preclinical mouse models. A second, unrelated Compound 22a is a 2-phenyl-3,4-dihydro-2H-benzo[f]chromen-3-amine derivative acting as a potent dipeptidyl peptidase 4 (DPP-4) inhibitor. This version is being investigated for the treatment of type 2 diabetes, showing significant glucose tolerance improvement and a long-acting pharmacokinetic profile comparable to omarigliptin in animal studies.
