Compound 250 is a synthetic derivative of the plant alkaloid securinine, developed as a differentiation therapy for acute myeloid leukemia (AML). It acts as a potent inhibitor of thioredoxin reductase (TrxR), a key redox mediator. By inhibiting TrxR, compound 250 induces oxidative metabolic stress and depletes mitochondrial spare respiratory capacity, which triggers the differentiation of leukemic myeloid progenitors into mature cells and induces cell death. It has demonstrated nanomolar potency in AML cell lines and patient samples, including those resistant to conventional chemotherapy like doxorubicin, and has shown efficacy in reducing leukemic burden in preclinical mouse models.
