Overview

Compound 26B is a small molecule inhibitor of phosphodiesterase 4D (PDE4D) that was identified and characterized as a potential therapeutic agent for various cancers. Research presented at the AACR 2013 meeting by Dechen Lin and colleagues (including researchers from the NIH Chemical Genomics Center and Cedars-Sinai Medical Center) demonstrated that PDE4D is frequently upregulated in solid tumors and promotes cancer cell proliferation. Compound 26B was shown to induce apoptosis and inhibit growth in multiple cancer cell lines, including melanoma, by increasing intracellular cAMP levels. This mechanism leads to the lineage-dependent induction of the pro-apoptotic protein BIM and the downregulation of the transcription factor MITF. The compound serves as a pharmacological tool to validate PDE4D as a targetable enzyme in oncology.
