Compound 27 is an optimized IRAK1/4/pan-FLT3 kinase inhibitor. It demonstrates potent and selective inhibition of Interleukin-1 receptor-associated kinase 1 (IRAK1), Interleukin-1 receptor-associated kinase 4 (IRAK4), and Fms-like tyrosine kinase 3 (FLT3). The drug candidate exhibits reduced hERG block and good pharmacokinetic properties. In preclinical studies, specifically a mouse xenograft model of acute myeloid leukemia (AML), compound 27 has shown superior survival prolongation compared to gilteritinib, an FDA-approved FLT3 inhibitor used for AML treatment.
