Compound 29a is a novel, liver-targeting phosphoric acid thyroid hormone receptor agonist. It was developed through the optimization of pharmacophores from existing THR-β agonists like VK2809 and resmetirom. Preclinical studies have shown that compound 29a effectively lowers cholesterol in a cholesterol-fed rat model and demonstrates potent anti-MASH effects in HFD-CCl4-induced mice MASH model. It exhibits favorable pharmacokinetic properties and a good safety profile, with primary distribution in liver and kidney tissues. These findings suggest that compound 29a holds considerable promise for the treatment of Metabolic dysfunction-associated steatohepatitis (MASH) and potentially other inflammatory and fibrotic diseases.
