Overview

Compound 4133 is a novel small molecule arylpiperazine-sulphonamide derivative being investigated for its neuroprotective potential in the treatment of Parkinson's disease. Developed by researchers at the University of Hertfordshire, the compound is part of a chemical series disclosed in patent WO 2016/051155. Compound 4133 is designed to address the underlying pathophysiology of Parkinson's by protecting dopaminergic neurons from mitochondrial dysfunction and proteasomal inhibition. In preclinical in vitro models using differentiated SH-SY5Y cells, compound 4133 demonstrated significant neuroprotective effects, improving cell viability by approximately 82.55% against MPP+-induced mitochondrial toxicity and 80% against lactacystin-induced protein clearance impairment. These findings suggest that the compound may counteract multiple pathogenic mechanisms simultaneously, offering potential as a disease-modifying therapy.
