Compound 47 is a synthetic benzylidene-indolinone derivative that acts as a multi-targeted tyrosine kinase inhibitor (TKI) for the treatment of hepatocellular carcinoma (HCC). Discovered through phenotypic screening for high HCC-specific cytotoxicity, the compound inhibits the phosphorylation of several receptor tyrosine kinases, most notably IGF-1R, Tyro3, and EphA2. In preclinical models, it has demonstrated the ability to suppress tumor growth, induce apoptosis, and inhibit cell migration, showing in vivo efficacy comparable to the standard-of-care TKI sorafenib. It also reduces the expression of the HCC biomarker alpha-fetoprotein (AFP), suggesting a targeted effect on oncogenic signaling pathways relevant to liver cancer.
