Compound 7b (also known as DC402234) is a potent, small-molecule peptidomimetic inhibitor of the SARS-CoV-2 main protease (Mpro or 3CLpro). Developed by the Shanghai Institute of Materia Medica (SIMM) and the Chinese Academy of Sciences, it was identified through structure-based drug design targeting the catalytic site of the protease, which is essential for viral replication. The molecule incorporates an aldehyde group that acts as a warhead, forming a reversible covalent bond with the Cys145 residue of Mpro. Preclinical studies demonstrated that Compound 7b possesses strong antiviral activity against SARS-CoV-2 in cell culture and shows favorable pharmacokinetic properties and safety in animal models. It was advanced into Phase 1 clinical trials as an intravenous formulation for the treatment of COVID-19.
