Drug intelligence / Profile preview
Compound 7l is a novel, dual-acting small molecule designed to simultaneously target peroxisome proliferator-activated receptor gamma (PPARγ) and histone deacetylases (HDACs), specifically HDAC4 and HDAC8. Developed through a collaborative effort involving Bharati Vidyapeeth's College of Pharmacy, the University of Texas at El Paso, the University of Napoli Federico II, the University of Applied Sciences Darmstadt, the University of Bari Aldo Moro, and the Institute of Hematology and Transfusion Medicine, compound 7l acts as a partial PPARγ agonist and a selective HDAC inhibitor. In preclinical studies, compound 7l has demonstrated significant anti-leukemic activity against chronic myeloid leukemia (CML) cells, particularly when combined with tyrosine kinase inhibitors (TKIs) like imatinib and dasatinib. The combination significantly reduces the clonogenic potential of CML stem-like cells, induces cell cycle arrest in the G0/G1 phase, and inhibits downstream MAPK, Ras, and PI3K-Akt signaling pathways without causing a significant increase in cell death, offering a promising therapeutic strategy to deplete quiescent leukemic stem cells and facilitate safe treatment-free remission.
