Compound 95 is a first-in-class dual inhibitor of butyrylcholinesterase (BChE) and p38α mitogen-activated protein kinase (p38α MAPK) developed for the treatment of Alzheimer's disease. Developed through structure-based drug design, it addresses two critical drivers of neurodegeneration: the cholinergic deficit and neuroinflammation. By inhibiting BChE, the compound aims to restore acetylcholine levels, while its inhibition of p38α MAPK targets the production of proinflammatory markers. Preclinical studies in mouse models of cognitive dysfunction (induced by scopolamine and lipopolysaccharide) have demonstrated that compound 95 significantly improves cognition and reduces neuroinflammatory markers, suggesting a comprehensive therapeutic strategy that targets both symptoms and underlying pathology.
