Compound gossypol acetate is a pharmaceutical formulation containing gossypol acetate, a polyphenolic compound derived from cottonseed, typically combined with vitamins such as Vitamin B6 (pyridoxine) to mitigate potential side effects. In oncology, gossypol acts as a pan-BCL-2 family inhibitor (BH3-mimetic), targeting pro-survival proteins including BCL-2, BCL-XL, and MCL-1 to promote apoptosis in malignant cells. Zhejiang Cancer Hospital is investigating this compound in a Phase II clinical trial as part of a triplet combination therapy with the CDK4/6 inhibitor dalpiciclib and the selective estrogen receptor degrader (SERD) fulvestrant. This combination is specifically aimed at treating advanced hormone receptor-positive (HR+), HER2-negative breast cancer in patients who have previously failed treatment with CDK4/6 inhibitors, leveraging gossypol's ability to overcome resistance by inducing programmed cell death.
