COMPOUND III is a newly synthesized indolylacetic derivative developed for the treatment of neurodegenerative diseases, particularly Amyotrophic Lateral Sclerosis (ALS). It was created by combining essential structural elements from opioids containing a 4-piperidinyl ring and indomethacin. Preclinical studies have demonstrated its ability to prevent the upregulation of tumor necrosis factor-alpha (TNF-α) in BV-2 microglial cells, preserve SHSY-5Y cell survival against neurotoxin exposure, and alleviate motor symptoms while improving survival rates in SOD1G93A ALS mouse models. These findings suggest its potential as a therapeutic agent for ALS by targeting neuroinflammation and promoting neuronal survival.
