Compound leucine granules are an oral amino acid formulation primarily composed of the branched-chain amino acids (BCAAs) L-leucine, L-isoleucine, and L-valine. The therapeutic rationale for its use in acute and chronic pancreatitis centers on the role of leucine as a potent nutrient signal that activates the mechanistic target of rapamycin (mTOR) pathway. Activation of mTOR promotes protein synthesis, inhibits autophagy-mediated protein degradation, and supports the maintenance of nutritional status in catabolic states. Additionally, the granules are being evaluated for their potential to modulate the gut microbiota and attenuate systemic inflammatory responses, thereby reducing postoperative complications and improving clinical outcomes in patients with pancreatic disorders.
