Compound3 is a novel small molecule inhibitor of the mitotic kinesin Eg5 (also known as KIF11), developed as a derivative of S-trityl-L-cysteine (STLC). It was synthesized and evaluated by researchers at the Shizuoka Cancer Center and the University of Shizuoka. Compound3 exerts its antineoplastic effects by disrupting the assembly of bipolar spindles during mitosis, leading to the formation of characteristic monoastral spindles and subsequent G2/M cell cycle arrest. Preclinical studies have demonstrated that Compound3 maintains potent growth-inhibitory activity in docetaxel-resistant non-small cell lung cancer (NSCLC) cell lines, such as PC-14/TXT, without showing cross-resistance to taxanes. This suggests its potential as a therapeutic candidate for patients with advanced NSCLC who have failed standard microtubule-stabilizing therapies.
