cON-05 is a potent and selective small molecule agonist of the G protein-coupled receptor 101 (GPR101), currently in preclinical development by Confo Therapeutics. GPR101 is an orphan GPCR that is highly expressed in the hypothalamus and pituitary gland, where it plays a critical role in the regulation of the growth hormone (GH) axis. Activation of GPR101 by cON-05 stimulates the secretion of growth hormone, making it a potential therapeutic candidate for the treatment of growth hormone deficiency (GHD) and other growth-related disorders. The drug was identified using Confo's proprietary ConfoBody technology, which stabilizes GPCRs in their active conformation to facilitate the discovery of high-affinity agonists.
