Overview

CON404 is a first-in-class, orally bioavailable small molecule agonist of the Vasopressin V2 receptor (V2R), currently in Phase 1 development for the treatment of Arginine Vasopressin Deficiency (AVP-D), formerly known as central diabetes insipidus. Developed by Confo Therapeutics using its proprietary G-protein coupled receptor (GPCR) targeting platform, CON404 is designed to mimic the activity of the antidiuretic hormone vasopressin. By selectively activating V2R in the kidneys, the drug promotes water reabsorption, thereby addressing the excessive thirst and polyuria associated with AVP-D. The compound was discovered through the use of ConfoBodies, which are nanobodies that stabilize GPCRs in their active state to facilitate the identification of potent small molecule agonists that target specific receptor conformations.
