conatumumab

Conatumumab is a fully human monoclonal antibody developed for the treatment of cancer. It acts as an agonist of tumor necrosis factor-related apoptosis-inducing ligand receptor 2 (TRAIL-R2, also known as death receptor 5 or DR5, TNFRSF10B). By binding to and activating TRAIL-R2/DR5 on tumor cells, conatumumab mimics the activity of native TRAIL, triggering caspase cascades that induce apoptosis in tumor cells. This mechanism selectively targets a variety of solid tumors and hematopoietic cancers with minimal toxicity to normal cells. Conatumumab was initially developed by Amgen and later licensed to Takeda for development in Japan; however, all clinical development has been discontinued.
