Conatumumab + FOLFIRI is a combination therapy composed of **conatumumab**, a fully human monoclonal antibody that acts as an agonist to **death receptor 5 (DR5, also known as TRAIL receptor 2)**, and **FOLFIRI**, a standard chemotherapy regimen including folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan. Conatumumab induces apoptosis selectively in tumor cells expressing DR5, while the FOLFIRI regimen disrupts DNA synthesis and replication through combined cytotoxic chemotherapy. This combination has been studied as a second-line treatment for patients with mutant KRAS metastatic colorectal cancer. In clinical studies, conatumumab + FOLFIRI was associated with a trend toward improved progression-free survival compared to FOLFIRI alone, and was generally tolerable, with acceptable toxicity profiles[1][3][5][7].
